Since discovery of bFGF (basic fibroblast growth factor, FGF-2) by Gospodarowicz in 1974 from bovine pituitary gland, human bFGF has been well characterized:
being a basic protein with pI 9.6 of MW ca. 17,000; h***ing high affinity to both heparin and heparan sulfate; and distributing a diversity of organs, tissues, and tumorous cells such as renal cell carcinoma. Recent investigation surrounding this protein has been focusing on relati***hips between diseases, especially, cancers and the concentration of bFGF in patient serum. Some h***e been discussed about co-relation between tumors and bFGF secretion. However, detection limits on most of the commercially ***ailable kits are inadequate compared with bFGF concentration in human serum. Replying to current demands, we h***e newly developed a highly sensitive bFGF ELISA Kit Wako utilizing multiple monoclonal antibodies well-characterized with human bFGF. The kit should enable the users to measure more than 0.25 pg human bFGF in serum and biological
samples by the standard ELISA method.

[Principle]
Based on enzyme-linked immunosorbant assay (ELISA) with sandwich-type format, the kit is designed to quantitate human bFGF in serum and biological fluid in the range of 5-500 pg/mL using multiple monoclonal antibodies well-characterized against human bFGF. Of 96-well microplate, all wells are coated with a mixture of two kind of monoclonal antibodies; clone No. 52 and 98 with similar antigenic recognition to strengthen the capture of antigen. By addition of specimens to antibody-coated wells, the antigen is specifically captured by the monoclonal antibodies. After washing out unbound serum components, HRP-conjugated Fab' prepared from monoclonal antibody, clone No. 3H3, h***ing another epitope recognition is administered in the wells, to form sandwich-type complexes of the antibodies and antigen on the well surfaces. The captured bFGF by the antibodies can be quantitated by the chromogenic reaction catalyzed by HRP conjugated with the Fab' fragment, using the standard curve made by human bFGF standa***upplied with the kit.
